Japan Ministry of Health, Labor and Welfare Approves Manufacturing and Marketing of Adtralza® (tralokinumab) in Japan for Adults with Atopic Dermatitis

BALLERUP, Denmark, 23, December 2022 – LEO Pharma A/S, a global leader in medical dermatology, announced today that the Japan Ministry of Health, Labor and Welfare (MHLW) has granted approval for the manufacturing and marketing of Adtralza® (tralokinumab) subcutaneous injection (S.C.) for adults with atopic dermatitis, which has inadequately responded to conventional therapies.

“Today’s approval is a significant advancement for those patients in Japan who continue to suffer from the often-debilitating symptoms of atopic dermatitis and are in need of a new treatment option to manage their chronic condition,” said Kook Hyun Son, General Manager of LEO Pharma K.K. “We have made substantial investments in our infrastructure, including the hiring of dedicated medical staff to ensure dermatologists receive the latest Adtralza clinical information to help inform treatment decisions for their patients.”

Adtralza is the first and only approved human, monoclonal antibody developed to specifically target and neutralize the IL-13 cytokine, which plays a key role in the immune and inflammatory process which are the underlying causes of atopic dermatitis signs and symptoms.1,2 The MHLW has approved Adtralza to be administered as a subcutaneous injection in adults with an initial dose of 600mg, followed by a 300mg dose given every other week.

About the ECZTRA 8 trial

ECZTRA 8 was a 16-week, double-blind, randomized, placebo-controlled phase 3 study in Japanese patients. The study evaluated the efficacy and safety of tralokinumab (300 mg) in combination with TCS in 106 adult patients with atopic dermatitis who were candidates for systemic therapy having an inadequate response to existing therapy.5

About the ECZTEND - long-term extension (LTE) trial

ECZTEND (Long-term Extension Trial in Subjects With Atopic Dermatitis Who Participated in Previous tralokinumab Trials) is an ongoing Phase 3, long-term five-year, open-label, single-arm

extension trial to evaluate the efficacy and safety of tralokinumab in patients with atopic dermatitis who participated in the previous tralokinumab monotherapy trials (ECZTRA 1 and ECZTRA 2), the combination therapy tralokinumab plus TCS trial (ECZTRA 3), the Drug-drug interaction (DDI) trial (ECZTRA 4), the vaccine trial (ECZTRA 5), the adolescent trial (ECZTRA 6), the oral cyclosporine A trial (ECZTRA 7), the combination therapy tralokinumab plus TCS trial in Japanese subjects (ECZTRA 8), and the tralokinumab monotherapy skin barrier function trial (TraSki). Patients were permitted to enter ECZTEND after completion of the parent trial regardless of their treatment response or whether they were treated with tralokinumab or placebo.6,7

About atopic dermatitis

Atopic dermatitis is a chronic, inflammatory, skin disease characterized by intense itch and eczematous lesions.8 Atopic dermatitis is the result of skin barrier dysfunction and immune dysregulation, leading to chronic inflammation.9 Type 2 cytokines, including IL-13, play a central role in the key aspects of atopic dermatitis pathophysiology.2

About Adtralza® (tralokinumab)

Tralokinumab is a human monoclonal antibody developed to specifically neutralize the IL-13 cytokine. Tralokinumab specifically binds to the IL-13 cytokine with high affinity, thereby inhibiting interaction with the IL-13 receptor α1 and α2 subunits (IL-13Rα1 and IL-13Rα2).1,2

Adtralza® / Adbry™ (tralokinumab-ldrm) is approved for the treatment of adults with moderate-to-severe AD in the European Union, United States, Great Britain, Canada, the United Arab Emirates, and Switzerland.

LEO Pharma presents new data on delgocitinib cream at EADV 2026 highlighting real-world outcomes in adults and new adolescent data in Chronic Hand Eczema (CHE)1.10.2026 12:00:00 CEST | Pressemeddelelse

Interim results from the German REACHED study provide the first real-world evidence on the effectiveness of delgocitinib cream in adults with moderate to severe Chronic Hand Eczema (CHE), with 28.6% of patients assessed as clear or almost clear after eight weeks of treatment, from none at baseline.1 New post hoc analyses from the phase 3 DELTA TEEN trial show greater reductions in daily HESD itch and pain scores with delgocitinib cream than with cream vehicle in adolescents aged 12-17 with moderate to severe CHE.2 In an oral presentation, new post hoc analyses from DELTA TEEN show that a higher proportion of adolescents reported no impact on quality of life at Week 16 with delgocitinib cream than with cream vehicle (27.0% vs 0%)3

New findings from a subgroup analysis of the 12-month TRACE study show that more than 70% of adults whose Atopic Dermatitis (AD) affected multiple high-burden areas – such as the head and neck, hands and feet, or genitals – had reduced involvement of those areas after 12 months of treatment with tralokinumab.1 New pre-specified and post hoc analyses from the phase 3 ADHAND trial show significantly greater improvements in quality of life, daily function and sleep at Week 16 with tralokinumab monotherapy than with placebo in adults with AD and moderate-to-severe hand involvement, with the two treatment groups separating within two weeks.2,3 In a post hoc analysis, adolescents aged 12-17 years treated across the phase 3 ECZTRA 6 trial and its ECZTEND extension, response rates remained high and stable through most of the extension period, and no new safety concerns were identified.5

The Committee for Medicinal Products for Human Use (CHMP) has recommended expanding the use of Anzupgo® (delgocitinib) cream to adolescents aged 12 to 17 years with moderate to severe chronic hand eczema for whom topical corticosteroids are inadequate or inappropriate. If approved by the European Commission, Anzupgo® would become the first therapy in the EU specifically approved for adolescents with moderate to severe chronic hand eczema, a patient population for whom there are currently no specifically approved treatment options. The positive CHMP opinion is supported by results from DELTA TEEN, a phase 3 trial investigating the efficacy and safety of Anzupgo® in adolescents with moderate to severe chronic hand eczema.1

The U.S. Food and Drug Administration (FDA) has accepted the New Drug Application (NDA) for dersimelagon for filing and granted it Priority Review, with a Prescription Drug User Fee Act (PDUFA) date expected by the end of February 2027. LEO Pharma has successfully closed the acquisition of dersimelagon from Tanabe Pharma, strengthening its late-stage pipeline and further leveraging its global dermatology platform. Dersimelagon is an investigational, once-daily oral melanocortin 1 receptor (MC1R) agonist for the treatment of erythropoietic protoporphyria (EPP) and X-linked protoporphyria (XLP), two rare genetic skin diseases with significant unmet medical need.
